AAA+ molecular machines play crucial roles in degrading abnormal proteins and reshaping proteomes as cells respond to environmental stress or pathogen attacks. The mechanisms underlying substrate specificity, and how these proteases adjust their activities in response to cellular and environmental changes, remain only partially understood. Our research focuses on unraveling the molecular mechanisms of substrate specificity and degradation by these molecular machines and their associated modulators, using structural biology, biochemistry, and cellular assays.
